    User Interface for Stand-alone Operation. The 212-LC is intended for use with Varian MS Systems under MS Workstation control. This pumping system is optimized for use in gradient or isocratic mode at flow rates below 2 mL/min.

    Unpacking and Installation The 212-LC System consists of two high pressure HPLC pumps. Pump A serves as the master control pump and will also be referred to as the pump which delivers solvent A. To distinguish the A and B pumps, the Serial Number for the A pump is labeled as AXXXX, whereas the B pump is labeled as BXXXX.

    Installing the Piston Wash Seals The 2 mL pump heads supplied with the Varian 212-LC pumps are capable of being configured for washing the back of the piston. This is important when pumping with a mobile phase containing a high concentration of buffer.

    Stand-alone Operation This section describes the use of the pump in Stand-alone mode, via the local keypads. Some of the information is also relevant to the operation under MS Workstation Control, in particular Priming, customizing the solvent configuration, and maintenance tasks.     (CF) for solvents other than those listed can be found below. Remote Control The 212-LC Pump System can be controlled remotely in one of two ways. External contact closures enable the system to be controlled by most HPLC autosamplers. For instructions see page 18. The 212-LC Pump System can also be controlled from a remote computer via the RS232 interface using MS Workstation version 6.6 SP1 and above.

    Determining Compressibility Factors Use the following procedure to calculate the compressibility factor for solvents not listed in the solvent library. In general, the more compressible the solvent, the higher the compressibility factor needs to be, for example water is 100, heptane is 200. 1.

    (T0) will display the time in 0.10 minute increments. The 212-LC system is now in the “Idle” condition and is ready to run the method file. The pump returns to “Idle” after running each method file.

    Methods are executed in the driver, and simple flow commands are sent to both pumps. The 212-LC MSWS Method can contain up to 100 steps specifying flow and composition, with a time resolution of 1 second. The method always executes a linear ramp of flow and composition from one step to the next.

    • Follow the on-screen instructions until the installation is complete. To verify that the 212-LC driver is present, right-click on the right side of the Workstation toolbar and select Enable/Disable Instrument Modules. In the resulting dialog, verify that the 212-LC driver is listed in the enabled or disabled modules.

    Push the new piston seal assembly over the end of the exposed ceramic piston with the fingertip or thumb, but do not allow your fingernails to contact the seal as damage may occur. This procedure aligns the piston seal assembly on the ceramic piston. It is normal for the seal not to push completely over the end of the ceramic piston.
